NB: just fixing a typo from previous review for biallelic and changing to amber for biallelic.

Only one family (not two) has been reported with AR inheritance, two homozygous siblings similarly affected with Lennox-Gastaut syndrome; their heterozygous siblings and mother are asymptomatic, father was not tested (Lammertse, H.C.A., et al. (2020)).

GoF has been suggested in this family for p.(Leu446Phe). Patch clamp demonstrated a two fold increase in synaptic transmission.
