Well-established gene-disease association. Loss of function is the mechanism of disease. Partial loss of fibrinogen causes dysfibrinogenemia/hypodysfibrinogenemia, while complete absence of fibrinogen (biallelic variants) causes afibrogenemia. There are also supporting mouse models that recapitulate the human phenotype.
